a) A cell requires the ability to sense and respond to its environment on a transcriptional level. List a mechanism as to how this might be achieved and an example process where this would be important?

b) Proteases are enzymes that degrade other proteins. Many proteases inside the cell are in the lysosome. Why do you think this is so and what properties, if any, of the lysosome contribute to regulation of protease activity inside the cell?

Answer #1

A) transcription factors activity in response to environmental signal is the primary response at the transcriptional level or gene expression level. Environmental signals activates transcription factors that binds with either enhancer of a gene loci to upregulate some genes or with repressor region to downregulate the expression of some genes. Combination of transcription factors are involved in the regulation in the transcriptional level. Once environemental signal sensed by the cells receptor it activates cascade of reactions to activate transcription factors that enter into the nucleus to upregulate or downregulate the gene expression

For example in lac operon of bacteria, the lactose source is identified and CRP levels gets high that activates the lac operon to synthesize lac genes.

B) lysosome are organs composed of more than 40 enzymes including proteases, lipase and nuclease to degrade or breakdown specific to each cellular components . Lysosomal proteases composed of aspartic protease, serine/cystein protease and each specific for different kind of proteins upon recognition

ALS is caused by the accumulation of misfolded protein that eventually leads to neuronal cell death. Just as is the case with prion diseases (e.g., Mad Cow disease), there is a spreading effect: even the presence of a small amount of misfolded (e.g., mutant) protein can cause other WT proteins of the same kind to become misfolded, which join the large misfolded aggregate forming in diseased cells. This is the principle mechanism thought to be behind the spreading of neurodegeneration...

Question 10-12 10. During the mating process, yeast cells respond to pheromones secreted by other yeast cells. These pheromones bind GPCRs on the surface of the responding cell and lead to the activation of proteins inside the cell. When a wild-type yeast cell senses the pheromone, its physiology changes in preparation for mating: the cell stops growing until it finds a mating partner. If yeast cells do not undergo the appropriate response after sensing a pheromone, they are considered sterile....

5.To gain entry, spikes of the SARS-CoV-2 bind to angiotensin-converting enzyme 2 (ACE2) found in epithelial cells especially of the respiratory and oral passageways. As such, these cells are susceptible to infection by SARS-CoV-2. ACE2 is a protein consisting of multiple alpha-helices that is embedded in, and spans, the membrane. What is this type of protein called, and what structure do the alpha helices represent? 6.Following binding of spike proteins to ACE2, SARS-CoV-2 likely gains entry into the cell either...
