1. Different mutations can either destroy pre-existing sites or create additional restriction sites which can result in the loss of a RFLP.

2. Positive YAC clones obtained from screening the library and identified by PCR or colony hybridization.

3. BamH and SnaB1 cleave YAC circularly as BamH1 cuts at telomeres and SnaB1 in middle of SUP4 selectable marker.
