You can use all three types of chromatography columns (ion exchange,affinity, gel filtration) in various combinations with one another to achieve a highly purified protein sample. Of the three, which would you use last in a sequence of various columns? Explain.

We should use gel filtration on top of gel because it will pass the particles on the basis of size. Then ion exchange column is placed because it traps the other charge particles and pass desired ones. At bottom of gel, we should us affinity column because it will bind all the desired particles. In this column we used specific antibody against our desired protein due to which we can extract our binding more. As in upper layer ion exchanges and gel filtration are used, there will be very high chances that affinity chromatography works in a high efficient manner and leads to extraction of desired substrate.
