What hypothetical mutation could you make to any mechanism so that when a cell replicates its DNA, the cell will die after? Think of two mutations. This is just 100% hypothetical and you could make up any mutation to any mechanism in replication, such as a beta clamp not working or something along those lines.

So we can make n number of mutation to make cell die :)

1. When the replication of DNA start and enzyme helicase opens the DNA, there is a mutation in the helicase enzyme which cause it to open the whole DNA (not the particular segment) which causes high tension in the structure of DNA leading to breaking up of its structure.

2. DNA polymerase enzyme instead of adding new nucleotides bases, start removing bases from the parent strand again leading to destruction in structure of DNA and ultimately cell dies.

3. When DNA replication is getting completed (in the end) when DNA ligase join the fragments, it joins the wrong fragments with each other leading to instability in DNA and mis-match information. Ultimately leading to death of organisim.
