In Cell culture is required cells are grown under laboratory conditions, which means outside the natural environment. Cells that are known as explants (in case of micropropagation of plant tissue )can be isolated from desired tissues for ex vivo culture in different manners. that's why Cells are grown and maintained at a particular temperature and gas mixture that is 37 °C, 5% CO2 required for mammalian cells.

so option D will be the correct answer.
