Mammalian SDH is present in 2 forms, one is activated form and another one is inactivated form. Transition from inactivated to activated state is initiated by binding of either substrate or competitive inhibitor or could be temperature dependent. So malonate here work as a activator for the SDH and its activity increases in the presence of malonate. Malonate binding to SDH is very strong and irreversible, hence SDH activity remains elevated in the presence of Malonate. Binding of Malonate to SDH causes conformational changes which convert SDH from less active state to high active state.
This malonate mediated activation has been observed in many animals like beef, Cow, rat, pig and yeast and occurs only in aerobic organism
