This is a typical profile of gel filtration chromatography. The higher molecular weight protein elutes first. Thus lower the elution volume of a protein, higher is its molecular weight. A standard graph of proteins with known molecular weight with the elution volume is plotted onto which the unknown protein can be mapped to find its relative molecular mass.
