*Spread plate technique involves using a sterilized spreader with a smooth surface made of metal or glass to apply a small amount of bacterial suspended in a solution over a plate . Dilution series from a sample are made and from this the upper limit of volume that can be taken to inoculate is 0.1 ml from the appropriate desired dilutiuon series on the centre of the surface of agar plate/spread plate.
