Hot spots are regions in proteins which are prone to mutations. Thus, over time, over evolution, such sites have undergone numerous mutation. Non-hotspots are generally termed as cold spots. Ht spots thus indicate areas, crucial for function or regulation, such that their mutation is often an adaptation for better survival. The improved function or the modified regulation ensures that the protein is better suited with the changing environment during evolution. Cold spots on the other side either don’t mutate or have mutated to a very similar residue in its place. These mutations are expected not to affect the protein structure or function to a significant impact.
