siRNAs are usually cut by DICER into 25 nucleotides long. There is mutation in DICER causing it to cut siRNAs 2 nucleotides long? Would this have an effect on mRNA silencing? Would this have an effect on the pathway of RNAi?

SiRNA, in the cell bind with other proteins and form RISC complex. Once this siRNA binds to its complementary mRNA leads to its degradation. This cause degradation of mRNA and no translation of protein. Thus silencing of gene.

If there is mutation in DICER , which lead to cut siRNA in 2 nucleotide London. In this condition sirna formed is very short length and is not able to carry out degradation of mRNA. Secondary, 20 to 25 bp of siRNA very specific to target gene. But in case of 2 long or can bind to non specifically at varios position. Because two bp will not be specific as 20 bp long sirna.

Main effect will be

1) nonspecific binding of sirna to various mrna.

2) sue to very short length it will be not able to carry out its function.
