The steps would be:

  1. Ribosomes that participate in the polypeptide synthesis are usually targeted to bind to the ER membrane by the presence of an amino acid sequence of the polypeptide chain that is being synthesized.
  2. This amino acid sequence is known as ER signal sequence which is present at the  N-terminus of the growing polypeptide chain.
  3. These signal sequences contain short stretches of hydrophobic amino acids that direct newly formed polypeptides to RER and simultaneously cleaved from the polypeptide chain as it transfer in to RER lumen.
