G protein- coupled receptor(GPCR) constitute a large protein family of receptors that detect molecules outside the cell and activate internal signal transduction pathways and cellular responses. While coupling with G protein they pass through the cell membrane seven times and thus, they are also known as seven transmembrane receptors.

Signaling of GPCRs is terminated by

1. Phospohorylation of receptors by cAMP activated PKA, which blocks the interaction with G- protein.

2. Hydrolysis of GTP to GDP by G-protein alpha.

Cleavage of GPCRs by receptor specific serine or threonine proteases regulates signal transduction and this mechanism is not involved in the termination process.
