A point mutation within the exonic functional part of DNA sequence would be most likely result in the production of a protein that differs from the non-mutated form by only a single amino acid, because exons are the coding nucleotide sequences of DNA that code for the synthesis of amino acids in a protein.

If a mutation prevents transcription then m-RNA would not be formed in the nucleus and a mutation that prevents m-RNA from exiting the nucleus to cytoplasm would lead to stop the process of translation and finally proteins never be synthesized in the cytoplasm of cell.
