Why is MNase used for the isolation of DNA bound to histones?
The digestion of chromatin is time-consuming than that of protein-free DNA. Micrococcal nuclease (MNase) can be used to digest the DNA bound with histones. This enzyme promotes dsDNA breaks within nucleosomal linker regions as an endonuclease. It is also reported to reduce the size of the chromatin fragment by exonucleolytic digestion.
Why is alkaline SDS used for plasmid DNA isolation but not for genomic DNA isolation?
-When DNA is DNA coiling is performed by the presence of coiled transcription is histones, inhibited chromatin, inhibited chromatin, activated histones, activated
