The digestion of chromatin is time-consuming than that of protein-free DNA. Micrococcal nuclease (MNase) can be used to digest the DNA bound with histones. This enzyme promotes dsDNA breaks within nucleosomal linker regions as an endonuclease. It is also reported to reduce the size of the chromatin fragment by exonucleolytic digestion.
