A) From the video 'setting up the API20E', the sample was collected from hospital sample which may contain mixed of microorganism. The pure culture plate is labeled as UTI which means sample collected from patient with urinary tract infections. Most probably the sample may be midstream urine or urithral discharge. The pure culture plate have many isolated colony. The each isolated colony indicate a single type of bacteria. And it is carefully picked up by using a sterile inoculation loop and mixed on to sterile distilled water for caring out API20E test.

B) the inoculation loop is flame sterilised before use to privent contamination with other bacterial species. If any other bacterial species present in the loop it may lead to false results in the API20E test. And interfere with the identification of the bacteria.

C) the API20E test strip is incubated at 37 degree celsius for 18 to 24 hour's.
