For the production of transgenic E coli -

1. Isolate gene encoding for insulin in human beings using restriction digestion.

2. Isolate plasmid from E coli. It should have an antibiotic resistance gene (say, ampR), origin of replication and polylinker site.

3. Cut the plasmid at polylinker site using the same restriction enzyme which was used to cut insulin gene from human genome.

4. Insert CaMV35 promoter into the plasmid. This will lead to an increased expression of gene encoding for insulin and this will lead to more production of insulin.

5. Insert the gene into the plasmid and this will lead to the formation of Recombinant DNA molecule.

6. Introduce this Recombinant DNA molecule into E coli.

7. Let it reproduce and produce insulin.

8. Isolate the insulin produced by this transgenic E coli.
