Southern blotting can be used while dealing with plasmid DNA. In this case plasmid to plasmid hybridization, a type of southern blotting is performed. In the first step, plasmid DNA is digested with proper restriction enzyme and then fragments are put onto gel electrophoresis and then blotting is done. Then a fragment from the gel or a cloned plasmid fragment is used as a molecular or hybridization probe. Yes, we use the plasmid as a molecular probe.
